摘要:enterohemorrhagicescherichiacolio157,averocytotoxin(vt1/2)-producingpathogen,canbedeadlybecauseitcaninduceacuteorchronicrenalfailure.tospeeduptheclinicaldiagnosisofrelatedsyndromescausedbye.colio157,thereisanurgentneedforrapid,simple,andreliableanalyticaltoolsforitsquantitation.inthisstudy,wedevelopedanovelelectrochemicalcompetitivegenosensor,featuringgold-electrodepositedscreen-printedelectrodes(nanoau/spe)modifiedwithaself-assembledmonolayerofthiol-cappedsingle-strandeddna(captureprobe),forthedetectionoftherfbegene,whichisspecifictoe.colio157.thisassayfunctionsbasedoncompetitionbetweenthetargetgene(complementarytothecaptureprobedna)andreporterdna-tagged,hexaammineruthenium(iii)chloride-encapsulatedliposomes.thecurrentsignalofthereleasedliposomalru(nh3)63+wasmeasuredusingsquarewavevoltammetry,yieldingasigmoidallyshapeddose−responsecurvewhoselinearportionwasovertherangefrom1to106fmol.thisliposomalcompetitiveassayprovidesanamplificationrouteforthedetectionoftherfbegeneatultratracelevels;indeed,wecoulddetectaslittleas0.75amolofthetargetrfbedna(equivalenttotheamountpresentin5μlofa0.15pmsolution).
