背景:metaloxidenanoparticles(nps)havebeenwidelyusedinindustry,cosmetics,andbiomedicine.
目的:weexaminedhazardsofseveralwell-characterizedhighproductionvolumenpsbecauseofincreasingconcernaboutoccupationalexposureviainhalation.
方法:apanelofwell-characterizednps[ceriumoxide(ceo2np),titaniumdioxide(tio2np),carbonblack(cbnp),silicondioxide(sio2np),nickeloxide(nionp),zincoxide(znonp),copperoxide(cuonp),andamine-modifiedpolystyrenebeads]wasinstilledintolungsofrats.weevaluatedtheinflammationpotenciesofthesenps24hrand4weekspostinstillation.fornpsthatcausedsignificantinflammationat24hr,wetheninvestigatedthecharacteristicsoftheinflammation.allexposureswerecarriedoutatequal-surface-areadoses.
结果:onlyceo2np,nionp,znonp,andcuonpwereinflammogenictothelungsofratsatthehighdosesused.strikingly,eachoftheseinducedauniqueinflammatoryfootprintbothacuy(24hr)andchronically(4weeks).acuy,patternsofneutrophilandeosinophilinfiltratesdifferedafterceo2np,nionp,znonp,andcuonptreatment.chronicinflammatoryresponsesalsodifferedafter4weeks,withneutrophilic,neutrophilic/lymphocytic,eosinophilic/fibrotic/granulomatous,andfibrotic/granulomatousinflammationbeingcausedrespectivelybyceo2np,nionp,znonp,andcuonp.
结论:differenttypesofinflammationimplydifferenthazardsintermsofpathology,risks,andriskseverity.invitrotestingcouldnothavedifferentiatedthesecomplexhazardoutcomes,andthishasimportantimplicationsfortheglobalstrategyfornphazardassessment.ourresultsdemonstratethatnpscannotbeviewedasasinglehazardentityandthatriskassessmentshouldbeperformedseparayandwithcautionfordifferentnps.
