摘要:poordrugencapsulationefficiencyandrapidreleaseoftheencapsulateddruglimittheuseofnanoparticlesinbiomedicalapplicationsinvolvingwater-solubledrugs.wehavedevelopedanovelpolymer-surfactantnanoparticleformulation,usingtheanionicsurfactantaerosolottm(aot)andpolysaccharidepolymeralginate,forsustainedreleaseofwater-solubledrugs.particlesizeofnanoparticles,asdeterminedbyatomicforcemicroscopyandtransmissionelectronmicroscopy,wasintherangeof40–70nm.weaklybasicmoleculeslikemethyleneblue,doxorubicin,rhodamine,verapamil,andclonidinecouldbeencapsulatedefficientlyinaot-alginatenanoparticles.invitroreleasestudieswithbasicdrugmoleculesindicatethatnanoparticlesreleased60–70%oftheencapsulateddrugover4weeks,withnearzero-orderreleaseduringthefirst15days.studieswithanionicdrugmoleculesdemonstratepoorerdrugencapsulationefficiencyandmorerapiddrugreleasethanthoseobservedwithbasicdrugs.furtherstudiesinvestigatingtheeffectofsodiumconcentrationinthereleasemediumandthechargeofthedrugsuggestthatcalcium-sodiumexchangebetweennanoparticlematrixandreleasemediumandelectrostaticinteractionbetweendrugandnanoparticlematrixareimportantdeterminantsofdrugrelease.inconclusion,wehaveformulatedanovelsurfactantpolymerdrugdeliverycarrierdemonstratingsustainedreleaseofwater-solubledrugs.
