摘要:photodynamictherapy(pdt)isapromisingtreatmentmodalityforcancer.pdtisbasedontheconceptthatphotosensitizers,whenexposedtolightofspecificwavelength,generatecytotoxicreactiveoxygenspecies(ros)capableofkillingtumorcells.theeffectivenessofpdthasbeenlimitedinpartbythelackofphotosensitizersthataccumulatesufficientlyintumorcellsandpooryieldofrosfromexistingphotosensitizers.inthisreport,weinvestigatedwhetheraerosolot-alginatenanoparticlescanbeusedasacarriertoenhancethetherapeuticefficacyofamodelphotosensitizer,methyleneblue.methyleneblueloadednanoparticleswereevaluatedforpdteffectivenessintwocancercelllines,mcf-7and4t1.encapsulationofmethyleneblueinnanoparticlessignificantlyenhancedintracellularrosproduction,andtheoverallcytotoxicityfollowingpdt.italsoresultedinhigherincidenceofnecrosis.greatereffectivenessofnanoparticlescouldbecorrelatedwithhigheryieldofroswithnanoparticle-encapsulatedmethyleneblue.further,treatmentoftumorcellswithnanoparticle-encapsulatedmethyleneblueresultedinsignificantnuclearlocalizationofmethylenebluewhilefreedrugtreatmentresultedinitsaccumulationmainlyintheendolysosomalvesicles.inconclusion,encapsulationofmethyleneblueinaerosolot-alginatenanoparticlesenhanceditsanticancerphotodynamicefficacyinvitro.increasedrosproductionandfavorablealterationinthesubcellulardistributioncontributetotheenhancedpdtefficacyofnanoparticle-encapsulatedphotosensitizer.
